In my opinion, the most important aspect is the title tag, the keyword near to the left would be most relevant to your content and if your content is related to that particular keyword, you will get good ranking for that keyword.

Well, in optimizing a keywords, keywords should be relevant to your site niche. It would be better if you optimize the long-tail keywords first, since short-tail is more competitive than long-tail. For better keyword suggestions, use Google Adwords. It provides better keywords that you can use for your site.
